1. Is it possible to manually draw in the markers or arrows to slice without the need for an automatic marker with sensitivity? If so how?
2. everytime we load a sample,does it have to default to stretch,or can it be defaulted to play back in other preferences? such as sample/slice/granular?

I hate this about UVI workstation too. It should not default to stretch when you load a sample. At least let it be a preference. I usually don't want to use Stretch and when I do I would set it manually. In UVI workstation it will even override this setting to stretch if you load a sample and turn it off the loading another sample sets it back. Loading a sample should not change any parameters.

In that case why don't you DnD on the mapping area ?
In that case it defaults to traditional sample playback.
It loads in stretch only if you DnD directly on the part.

I don't have M53 yet. Just I have the same issue with UVI. Either way, I don' like using D&D much it is a bit awkward (RSI). I normally double click to load or enter after using arrows on my keyboard. Also use the arrows on the part to increment to next sample. I assume this is the same with M53?
