I am running DP 4.61.
Currently I am working to sequence a large score. For my work flow I have placed markers to represent the form of the entire score. But because I am varying the tempo to play various sections in accurately, the markers are moving in their location relative to bar numbers. This is not how I want to work! I know that locked markers are locked to SMTPE time and not measure numbers. But I would like to know if there is way to have markers (or an equivalent) locked to measure numbers so that I can get to places in the sequence and be assured that it is the correct measure number ... while varying the tempo!

If you 'unlock' the markers, they will actually be locked to the measures & beats. As you are probably aware, it's impossible for a marker to be locked to both SMPTE & Measure position (ticks).
Personally I think it's bad terminology on DP's part. It's all about what the markers are 'locked' to. Either MIDI ticks or Absolute position. This is where the Pro Tools system works well - although if you screw up in PT and create the wrong type of markers, you have to go through them one at a time to change them (Aaaaaargh!).

It would be cool if DP had different types of markers that could be color coded. So, one color could connote a marker locked to SMPTE and another color a marker locked to mensural time. Then, you wouldn't actually need to engage a lock at all for any given marker. Each would have only one state true to its purpose.

Yes, and add different key commands depending on which type we want to create, and we'd be flying.
