(1) How can I make a sample play till finished instead of running the length of MIDI down?

When you select the sample, on the left side of the main window you can select Loop to be One-shot (or something like that). That'll do it.

Poke around in the window in MachFive, there's a lot in there that's pretty self-explanatory and well worth exploring.
