This happens in Cubase 5.1, but did not used to happen in earlier versions from Cubase 4 backwards, in which MachFive always played as soon as a project was opened. Now it is necessary to 'trick' the samples into playing by reloading one or more until one kicks in, and then all of them play for the remainder of the session. Usually this happens the first time, but sometimes it can take dozens of trick loads to provoke MachFive into playing its samples.
I have tested the same plug-in with Digital Performer 6.02, and it plays first time, as you'd expect I suppose, given that DP is also MOTU's product, however I have also tested it with Sibelius 6.01 and it plays just fine there too. Cubase 5 is the only host DAW which causes MachFive to not play like this.
I have repaired permissions. I have also raised support tickets also with Mark of the Unicorn and with Steinberg and will report back anything they tell me. I had a lengthy phone support with MOTU and they were unable to shed light after establising the MachFive workds just fine with MOTU apps, like DP.
Anyone else had issues like this, and if so, a solution?

So MOTU tech didn't offer any suggestions?
What comes to mind is to:
1. Try reinstalling Mach Five, maybe just the VST version if the installer allows this.
2. Maybe try pulling your the "com.motu.MachFive2.plist located in the "user/Library/Preferences" onto your desktop, then restart Cubase, open a project with M5. This should create a new plist.
