I come from a visual arts background. I've spent the majority of my life cultivating skills as an illustrator, not a musician. My 2¢: Quantization is your enemy. Avoid using the same samples in duplicated instruments. I've also had good results with slight detuning, however I have had many a brickbat thrown my way for even suggesting such a thing. A slight vibrato is well within acceptable limits of detuning.
