Update. MOTU instructed to pull the 424 card _and then_ re-install the drivers without the card installed. Although this fix has worked for some, my 8 core Mac still 'permanently' loses the 2408's after falling asleep.
Any help would be great...

Hi Frank, I didn't see your post until after I submitted mine. Looks like we both have the same problem. Our only similarities in hardware/software is OSX 10.4.10 and PCI Audio 1.2.0.
