From the MOTU site:

"If your computer uses standard PCI (5V) or PCI-X (3.3V) slots, our PCIX-424 card will be compatible with your system."

Last year I bought a new 2408mk3 which came with the PCIX-424 card. At first the card fit and worked just fine in my G4 MDD DP 1.25GHz - then recently I upgraded to a PCI-X G5, and the card works fine in the new computer as well.

You should be all set. Your only problem is: if you ever want to get a new-ish computer, that computer will have PCI-e slots and you'll have to upgrade the MOTU card to the PCIe-424.

I also have a G4 with a 424 system, and it's worked well for some time. I'm having a few problems now, which is why I'm here, but it's been rock solid for a couple of years of heavy use.
