Turn on audible mode (the speaker in the transport window) and you'll hear MIDI notes when you click on them.

Try using DP's 'wait' function. Playback can be started from a sustain pedal, from a MIDI keyboard, or any MIDI activity.

Take a look at (and demo) the McDSP 6030 Ultimate Comp. Excellent LA2, 1176, DBX Over Easy, Vari-Mu models and more. My go-to comp plugin. Easy to dial in and all the models sound great!

AFAIK, the PCI-424e doesn't operate any faster than 1x? I need to check, but I'd bet that it makes absolutely no difference if you're in the 4x slot or the 16x slot when you're dealing with audio. You may want to try reinserting into the bottom slot as I believe the bottom slot is 16X, the two PCIe slots above being bandwidth limited to 4x.
